Variantes germinales en pacientes con cáncer gástrico esporádico del suroccidente colombiano

Introducción: el cáncer gástrico (CG) es un problema de salud pública a nivel mundial, y Colombia es una de las regiones con mayor incidencia de esta enfermedad. Diversos factores explican el desarrollo del CG, entre ellos las mutaciones germinales (enfermedad hereditaria), que contribuyen al riesgo individual de padecer esta patología.
Objetivo: identificar variantes germinales en el exoma de pacientes diagnosticados con CG esporádico procedentes de regiones de alta y baja incidencia en Colombia.
Materiales y métodos: se recolectaron cuatro muestras de sangre de pacientes con CG del departamento de Nariño, Colombia, para secuenciación de exoma completo (WES). El análisis bioinformático incluyó el control de calidad de los datos crudos; el alineamiento de las lecturas contra el genoma humano de referencia; el llamado de variantes; la anotación, filtración y priorización de variantes; la visualización de datos, y el análisis de vías funcionales de los genes con variantes deletéreas.
Resultados: en total se identificaron 1.800.145 variantes germinales en los pacientes analizados. Cinco de estas variantes se reportaron en ClinVar con significancia clínica de patogenicidad, y estas alteraciones genéticas no han sido previamente descritas en pacientes con CG. Las variantes se encontraron en los genes ACADS (rs57443665), BRCA1 (rs55688530), FLNC (rs200502811), MCCC2 (rs757052602) y ACSS2 (rs59088485), que han sido previamente reportados y estudiados por el Atlas del Genoma del Cáncer (TCGA).
Conclusiones: se reportaron variantes moleculares raras y patogénicas relacionadas con la susceptibilidad al desarrollo de CG. Se invita a la comunidad académica a realizar estudios que permitan validar estos resultados.
